WHAT IT IS

A GLP-1 analogue that targets satiety and gastric emptying.

Semaglutide is a synthetic peptide that mimics GLP-1 (glucagon-like peptide-1), a hormone the gut releases after a meal. It is the same molecule sold under Ozempic (type-2 diabetes) and Wegovy (weight management) — the active ingredient is identical; the dosing and approved indication differ.

Its effect on weight comes from three combined actions: it increases the feeling of fullness after eating, slows gastric emptying, and modulates the insulin/glucagon response to glucose.

HOW IT WORKS

Mechanism of action and why food intake decreases spontaneously.

Semaglutide binds to GLP-1 receptors in the pancreas, the digestive tract and brain areas that regulate appetite (hypothalamus and brainstem). That binding produces three clinically measurable effects:

  • Satiety: patients describe getting full earlier and staying full longer, which reduces intake without the need for calorie counting.
  • Slowed gastric emptying: food stays in the stomach longer, which also explains the mild nausea reported in the first month.
  • Glycaemic regulation: it increases insulin secretion under hyperglycaemia and suppresses glucagon, improving blood-sugar control.

The effect on weight unfolds gradually over months; phase-III clinical trials report an average sustained loss of 10–15 % of starting weight when combined with diet and physical-activity changes.

WHO IS A CANDIDATE

The EMA indication in weight management is for adults with BMI ≥ 30 kg/m² (obesity), or ≥ 27 kg/m² (overweight) with at least one weight-related comorbidity (hypertension, type-2 diabetes, dyslipidaemia, sleep apnoea).

The licensed doctor reviews your questionnaire to verify the indication, rule out contraindications (personal or family history of medullary thyroid carcinoma, MEN-2, prior pancreatitis, pregnancy, breastfeeding) and check for interactions with your current medication before prescribing.

SAFETY

Side effects, contraindications and monitoring.
